HLA b-27 by Flow Cytometry Will reflex to HLA-B27 by PCR for confirmation when result is indeterminate.
Methodology
Flow cytometry
Rejection Criteria
Samples collected in lithium heparin tube Hemolysis
Reference Range(s)
Not present Will reflex to HLA-B27 by PCR for confirmation when result is indeterminate.
Specimen Requirements
Preferred Specimen
EDTA (lavender top) tube
Minimum Volume
2 mL whole blood
Sample Stability
3 days room temperature Note: refrigerated samples are unacceptable; however samples refrigerated for up to 72 hours will be tested and results individually assessed by a pathologist. Results may be rejected, requiring sample recollection.
Transport Temperature
Critical Room Temperature
Collection Instructions
Fill EDTA tube and gently mix by inverting the tube 8-10 times. Critical Room Temperature.
Alternative Specimen
4 mL sodium heparin (green top) whole blood. Critical Room Temperature (CRT). Note: confirmatory testing cannot be performed with sodium heparin (green top) results obtained by flow cytometry.
